Lipid Metabolism- 64 Elongation of fatty acids Elongation of fatty acids occurs in the microsomes by fatty acid elongase system. The microsomal elongation system can elongate: Saturated fatty acids with 10 carbons or more. Unsaturated fatty acids. Elongation of palmitate to stearate: Palmitate is activated to palmitoyl-CoA. Palmitoyl-CoA condenses with malonyl-CoA. The β-ketoacid produced will be: reduced by NADPH+H+, dehydrated and reduced by NADPH+H+. The same rounds as in fatty acid synthesis and stearate will be produced. Regulation of elongation of fatty acids Fasting reduces elongation of fatty acids. Malonyl-CoA is a powerful inhibitor of acyl-CoA carnitine transferase, necessary for transport of fatty acids to the mitochondria to be β -oxidized. This leaves the synthesized fatty acids in the cytoplasm for esterification and synthesis of triacylglycerols. Elongation of stearyl-CoA in the brain increases rapidly during myelination to produce C 22-C24 fatty acids that are present in Sphingomyelins.
